Я попробовал pythonnet с pypy на Win10. Он строит отлично, но затем не удается импортировать clr. Сбой происходит после этого вызова на снимке экрана.

Скорее всего, у вас не установлена библиотека python clr. В командной строке/терминале введите pip install clr. Если вы используете Linux/OS X, sudo перед командой, чтобы избежать проблем с разрешениями. После установки clr попробуйте импортировать его в программу на python.
Я пытаюсь проверить с PyCharm, и результат установки работает хорошо. Возможно, у вас возникли проблемы с показами IDE на снимке экрана.
